NLRP7 Promotes Choriocarcinoma Growth and Progression through the Establishment of an Immunosuppressive Microenvironment

The inflammatory gene NLRP7 is the major responsible for recurrent complete hydatidiform moles (CHM), an abnormal pregnancy that can develop into gestational choriocarcinoma (CC). However, role of in development and immune tolerance CC has not been investigated. Three approaches were employed to define development: (i) a clinical study analyzed human placenta sera collected from women with normal pregnancies, CHM or CC; (ii) vitro investigated impact knockdown on tumor growth organization; (iii) vivo used two mouse models, including orthotopic model. circulating cytokines upregulated cells CC. In cells, functions inflammasome-independent manner promoted their proliferation 3D organization. Gravid mice placentas injected invalidated NLRP7, exhibited higher maternal response, developed smaller tumors, displayed less metastases.
